Destiny: Prison of Elders loot system explained

Destiny's PvE activity, the Prison of Elders, rewards you with loot in a number of different ways. So which ones can you grind, and which can you only do once per week?
There are two ways you can get loot in Prison of Elders. The first is the random drops you get after completing the run and entering the treasure room. The other is opening the chests in the same room.
